A primary assessment of the endophytic bacterial community in a xerophilous moss (Grimmia montana) using molecular method and cultivated isolates BJM
Liu,Xiao Lei; Liu,Su Lin; Liu,Min; Kong,Bi He; Liu,Lei; Li,Yan Hong.
Investigating the endophytic bacterial community in special moss species is fundamental to understanding the microbial-plant interactions and discovering the bacteria with stresses tolerance. Thus, the community structure of endophytic bacteria in the xerophilous moss Grimmia montana were estimated using a 16S rDNA library and traditional cultivation methods. In total, 212 sequences derived from the 16S rDNA library were used to assess the bacterial diversity. Sequence alignment showed that the endophytes were assigned to 54 genera in 4 phyla (Proteobacteria, Firmicutes, Actinobacteria and Cytophaga/Flexibacter/Bacteroids). Of them, the dominant phyla were Proteobacteria (45.9%) and Firmicutes (27.6%), the most abundant genera included Acinetobacter,...

Ambient pH-regulated enzime secretion in endophytic and pathogenic isolates of the fungal genus Colletotrichum Scientia Agricola
Maccheroni Jr.,Walter; Araújo,Welington Luiz; Azevedo,João Lúcio.
In fungi a genetic system ensures that enzymes are secreted mainly at ambient pH values corresponding to their optima of activity. Although a great deal of information has been obtained concerning this environmental response, there is a lack of studies involving phytopathogenic, endophytic and entomopathogenic fungi as well as different aspects of fungus-host interactions. This study compares in a plate-clearing assays, the effect of ambient pH in the secretion of amylase, cellulase, lipase, pectinase and protease by endophytic, phytopathogenic, and entomopathogenic isolates belonging to several species of Colletotrichum. All enzymes were secreted in a pH-dependent manner by all isolates. Endophytes and pathogens showed distinct patterns of protease...

Antagonistic endophytic bacteria associated with nodules of soybean (Glycine max L.) and plant growth-promoting properties BJM
Zhao,LongFei; Xu,YaJun; Lai,XinHe.
Abstract A total of 276 endophytic bacteria were isolated from the root nodules of soybean (Glycine max L.) grown in 14 sites in Henan Province, China. The inhibitory activity of these bacteria against pathogenic fungus Phytophthora sojae 01 was screened in vitro. Six strains with more than 63% inhibitory activities were further characterized through optical epifluorescence microscopic observation, sequencing, and phylogenetic analysis of 16S rRNA gene, potential plant growth-promoting properties analysis, and plant inoculation assay. On the basis of the phylogeny of 16S rRNA genes, the six endophytic antagonists were identified as belonging to five genera: Enterobacter, Acinetobacter, Pseudomonas, Ochrobactrum, and Bacillus. The strain Acinetobacter...

Biological control of phytopathogenic fungi by endophytic actinomycetes isolated from maize (Zea mays L.) BABT
Costa,Francisco Gheler; Zucchi,Tiago Domingues; Melo,Itamar Soares de.
This work aimed a survey on the biodiversity of maize endophytic actinomycete, and an evaluation of their potential to control the phytopathogenic fungi. From several regions of São Paulo state, 40 strains were isolated from the healthy maize plants. The identification of these strains, based on morphological properties and fatty acid methyl ester (FAME) profile showed that most of them belonged to the Streptomyces genus. These isolates were first screened for the growth inhibition of phytopathogenic fungi and results showed that all the isolate were able to inhibit the development of at least one tested pathogen. Two selected isolates were then evaluated for the control of P. aphanidermatum in cucumber (Cucumis sativa L.) under greenhouse conditions....

Dark septate endophytic pleosporalean genera from semiarid areas Naturalis
Knapp, D.G.; Kovács, G.M.; Zajta, E.; Groenwald, J.Z.; Crous, P.W..
Dark septate endophytes (DSE) are distributed worldwide as root-colonising fungi, and frequent in environments with strong abiotic stress. DSE is not a taxon, but constitutes numerous fungal taxa belonging to several orders of Ascomycota. In this study we investigate three unidentified DSE lineages belonging to Pleosporales that were found previously in semiarid sandy grasslands. For molecular phylogenetic studies seven loci (ITS, partial 18S nrRNA, 28S nrRNA, actin, calmodulin, transcription-elongation factor 1-α and ß-tubulin genes) were amplified and sequenced. Based on morphology and the resulting molecular phylogeny these isolates were found to represent three novel genera within the Pleosporales, namely Aquilomyces, Flavomyces and Darksidea, with eight...

Detection of siderophores in endophytic bacteria Methylobacterium spp. associated with Xylella fastidiosa subsp. pauca PAB
Lacava,Paulo Teixeira; Silva-Stenico,Maria Estela; Araújo,Welington Luiz; Simionato,Ana Valéria Colnaghi; Carrilho,Emanuel; Tsai,Siu Mui; Azevedo,João Lúcio.
The objective of this work was to study the production of siderophores by endophytic bacteria Methylobacterium spp., which occupy the same ecological niche as Xylella fastidiosa subsp. pauca (Xfp) in citrus plants. The siderophore production of Methylobacterium strains was tested according to chromeazurol agar assay test (CAS), Csáky test (hydroxamate-type) and Arnow test (catechol-type). In addition, the ability of Xfp to use siderophores, in vitro, produced by endophytic bacteria as source of iron, was evaluated. All 37 strains of Methylobacterium spp. tested were CAS-positive for siderophore production. Methylobacterium spp. produced hydroxamate-type, but not catechol-type siderophores. In vitro growth of Xfp was stimulated by the presence of...

EFFECTS OF FUNGICIDES ON ENDOPHYTIC FUNGI AND PHOTOSYNTHESIS IN SEEDLINGS OF A TROPICAL TREE, Guarea guidonia (Meliaceae) Acta biol.Colomb.
GAMBOA GAITÁN,MIGUEL A.; WEN,SHIYUN; FETCHER,NED; BAYMAN,PAUL.
Endophytes are microorganisms that live within healthy plant tissues, and include fungi and bacteria. They can be mutualists, comensals or even latent pathogens. Presence of these endosymbionts may affect host physiology, for example by consuming products of photosynthesis (endophytes are heterotrophs) or producing toxic metabolites. In this work two fungicides were used to eliminate fungal endophytes from seedlings of Guarea guidonia. Lightsaturated photosynthesis (Amax) was measured in endophytefree plants and compared with control plants. Each fungicide killed different fungal endosymbionts. Phomopsis was more susceptible to benomyl while Colletotrichum was more susceptible to propiconazole. Although suggestive, values of Amax were not significantly...

ENZYMATIC AND ANTAGONIST ACTIVITY OF ENDOPHYTIC FUNGI FROM Sapindus saponaria L. (SAPINDACEAE) Acta biol.Colomb.
SANTOS,Caroline Menicoze dos; RIBEIRO,Amanda da Silva; GARCIA,Adriana; POLLI,Andressa Domingos; POLONIO,Julio Cesar; AZEVEDO,João Lúcio; PAMPHILE,João Alencar.
ABSTRACT Endophyte microorganisms have great biotechnological interest, with features applicable to different areas and are potentially useful in agriculture. The current study determines the biotechnological potential of endophytic fungi, isolated from leaves of Sapindus saponaria, to control phytopathogenic fungi and evaluate their enzyme production. Molecular taxonomy was performed by sequencing of the ITS1-5.8S-ITS2 ribosomal DNA region, identifying the genera Phomopsis, Sordariomycetes, Diaporthe, and Colletotrichum. In vitro antagonism against phytopathogens showed better results against Fusarium solani and provided inhibition indices between 41.8 % and 67.5 %. The endophytic strain SS81 (Diaporthe citri) presented the highest antagonism index...

Exploring interactions of plant microbiomes Scientia Agricola
Andreote,Fernando Dini; Gumiere,Thiago; Durrer,Ademir.
A plethora of microbial cells is present in every gram of soil, and microbes are found extensively in plant and animal tissues. The mechanisms governed by microorganisms in the regulation of physiological processes of their hosts have been extensively studied in the light of recent findings on microbiomes. In plants, the components of these microbiomes may form distinct communities, such as those inhabiting the plant rhizosphere, the endosphere and the phyllosphere. In each of these niches, the "microbial tissue" is established by, and responds to, specific selective pressures. Although there is no clear picture of the overall role of the plant microbiome, there is substantial evidence that these communities are involved in disease control, enhance...

Fungal endophyte diversity in the leaves of the medicinal plant Myracrodruon urundeuva in a Brazilian dry tropical forest and their capacity to produce L-asparaginase Acta Botanica
Pádua,Ana Patrícia Sousa Lopes de; Freire,Karla Torres Lins de Sousa; Oliveira,Thays Gabrielle Lins de; Silva,Leticia Francisca da; Araújo-Magalhães,Gianne Rizzuto; Agamez-Montalvo,Gualberto Segundo; Silva,Iolanda Ramalho da; Bezerra,Jadson Diogo Pereira; Souza-Motta,Cristina Maria de.
ABSTRACT We studied the fungal endophyte community of the leaves of Myracrodruon urundeuva in Brazil along with their potential to produce L-asparaginase. In total, 187 endophytes were isolated. The colonization rate of leaf fragments from caatinga (10.41 %) was lower than those from brejo de altitude (39.58 %). Sequences of ITS rDNA from all endophytes revealed relationships with Ascomycota (Botryosphaeriales, Chaetothyriales, Diaporthales, Eurotiales, Glomerellales, Hypocreales, and Pleosporales) and Basidiomycota (Polyporales). The most frequent endophytes were members of the genus Diaporthe. Talaromyces was an indicator genus for caatinga while Phyllosticta, Diaporthe, and Colletotrichum were for brejo de altitude. The composition of endophytic fungi...

Genetic diversity and population densities of endophytic Bacillus spp. in yam plants Bragantia
Souza,Jorge Teodoro de; Santos,Ana Cláudia Oliveira Souza; Monteiro,Fernando Pereira; Batista,Mariah Rodrigues; Santos,Patrícia Oliveira dos; Carvalho,Larissa Ferreira; Corrêa,Elida Barbosa.
ABSTRACT This study was conducted as a first step in exploiting endophytic bacteria to improve yam production. Little information is available on population densities and genetic diversity of endophytic bacteria in yam cultivated in tropical regions. This study demonstrated that higher densities of total endophytic bacteria and Bacillus occur in the interior of roots and decrease as the sampled plant organ departs from soil. Bacillus represents approximately 1% of the total bacterial endophytic population. Random Amplified Polymorphic DNA (RAPD) analyses performed with 88 randomly-selected Bacillus isolates resulted in 28 groups, and sequence analyses of the 16S region of the ribosomal DNA of 24 isolates representing all major RAPD groups revealed that...

Genetic variability in the endophytic fungus Guignardia citricarpa isolated from citrus plants Genet. Mol. Biol.
Glienke-Blanco,Chirlei; Aguilar-Vildoso,Carlos Ivan; Vieira,Maria Lúcia Carneiro; Barroso,Paulo Augusto Vianna; Azevedo,João Lúcio.
During some phases of of their life-cycle endophytic fungi colonize plants asymptomatically being found most frequently inside the aerial part of plant tissues. After surface disinfection of apparently healthy leaves from three varieties of mandarin orange and one tangor, and after incubation on appropriate culture medium, 407 fungal isolates were obtained, giving a total infection frequency of 81%. No fungal growth was observed from disinfected seeds, indicating that fungi are probably not transmitted via seeds. Of the fungal isolates, 27% belonged to the genus Guignardia, with 12 isolates being identified as Guignardia citricarpa Kiely, which is described as a citrus pathogen. The isolates were variable in respect to the presence of sexual structures and...

The diversity of citrus endophytic bacteria and their interactions with Xylella fastidiosa and host plants Genet. Mol. Biol.
Azevedo,João Lúcio; Araújo,Welington Luiz; Lacava,Paulo Teixeira.
Abstract The bacterium Xylella fastidiosa is the causal agent of citrus variegated chlorosis (CVC) and has been associated with important losses in commercial orchards of all sweet orange [Citrus sinensis (L.)] cultivars. The development of this disease depends on the environmental conditions, including the endophytic microbial community associated with the host plant. Previous studies have shown that X. fastidiosa interacts with the endophytic community in xylem vessels as well as in the insect vector, resulting in a lower bacterial population and reduced CVC symptoms. The citrus endophytic bacterium Methylobacterium mesophilicum can trigger X. fastidiosa response in vitro, which results in reduced growth and induction of genes associated with energy...
